CYP2C9 gene is an important member of cytochrome P**0 proteins (CYP). It is involved in the hydroxylation metabolism of many drugs, among which warfarin, tolbutamide, and phenytoin are all drugs with a narrow therapeutic index. CYP2C9 * 2 (rs******3, C**0T) and CYP2C9 * 3 (rs******0, A***5C) decreased CYP2C9 activity. The frequency of CYP2C9 * 2 and CYP2C9 * 3 in the Chinese population is 0% and 3%, respectively. CYP2C9 genetic polymorphism leads to the change of its enzyme activity, which leads to the phenomenon of racial and individual differences in drug metabolism, and even leads to the occurrence of serious adverse drug reactions. VKORC1 gene encodes subunit 1 of vitamin K cycloreductase complex. Related research showed that the - ***9g > A (rs******1) polymorphism of the VKORC1 gene affected the expression of VKORC1 and the sensitivity of warfarin. CYP4F2 gene-encoded protein belongs to cytochrome P**0 superfamily. Some researches found that CYP4F*3 (G***7A, rs******2) polymorphism is related to the clinical dosage of warfarin.
In this kit, CYP2C9*3, VKORC1 (- ***9g > A), CYP4F2*3 gene polymorphism was detected by using the temperature difference of hybridization between probes and different alleles. Primers were designed to amplify the site to be tested, and then the melt curve of the hybridization between the fluorescent-labeled specific probe and the amplified product was used to determine the genotype of the sample at the site. Different genotypes have different peak maps and Tm values so that qualitative analysis of detection results can be achieved.
